Glycolytic activity is required for the onset of neural plate folding during neural tube closure in mouse embryos

چکیده
Physiological hypoxia is critical for placental mammalian development. However, the underlying mechanisms by which regulates embryonic development remain unclear. We discovered that expression of glycolytic genes partially depends on in neuroepithelial cells E8.25 mouse embryos. Consistent with this finding, inhibiting glycolysis during early phase neural tube closure (E8.0–8.5) resulted a defect. In contrast, electron transport chain did not affect formation. Furthermore, affected cell proliferation, but differentiation and survival. Inhibiting repressed phosphorylation myosin light 2, consequent plate folding. Our findings revealed anaerobic proliferation apical constriction closure.

Biomechanical coupling facilitates spinal neural tube closure in mouse embryos.
Neural tube (NT) formation in the spinal region of the mammalian embryo involves a wave of "zippering" that passes down the elongating spinal axis, uniting the neural fold tips in the dorsal midline. Failure of this closure process leads to open spina bifida, a common cause of severe neurologic disability in humans. Here, we combined a tissue-level strain-mapping workflow with laser ablation of...
متن کاملIs programmed cell death required for neural tube closure?
Programmed cell death (PCD) plays an important part in animal development. It is responsible for eliminating the cells between developing digits, for example, and is involved in hollowing out solid structures to create cavities (reviewed in [1] [2]). There are many cases, however, where PCD occurs in developing tissues but its function is unknown. Important examples are seen during the folding,...
متن کاملApoptosis is not required for mammalian neural tube closure.
Apoptotic cell death occurs in many tissues during embryonic development and appears to be essential for processes including digit formation and cardiac outflow tract remodeling. Studies in the chick suggest a requirement for apoptosis during neurulation, because inhibition of caspase activity was found to prevent neural tube closure. In mice, excessive apoptosis occurs in association with fail...
متن کاملBmp2 is required for cephalic neural tube closure in the mouse.
BMPs have been shown to play a role in neural tube development particularly as dorsalizing factors. To explore the possibility that BMP2 could play a role in the developing neural tube (NT) beyond the lethality of Bmp2 null embryos, we created Bmp2 chimeras from Bmp2 null ES cells and WT blastocysts. Analysis of Bmp2 chimeras reveals NT defects at day 9.5 (E9.5). We found that exclusion of Bmp2...
متن کاملLoss of Gcn5 acetyltransferase activity leads to neural tube closure defects and exencephaly in mouse embryos.
Gcn5 was the first transcription-related histone acetyltransferase (HAT) to be identified. However, the functions of this enzyme in mammalian cells remain poorly defined. Deletion of Gcn5 in mice leads to early embryonic lethality with increased apoptosis in mesodermal lineages. Here we show that deletion of p53 allows Gcn5(-/-) embryos to survive longer, but Gcn5(-/-) p53(-/-) embryos still di...
